Hey Chris,
I also was getting "Access is denied" when trying to update the firmware. In one of the tabs, in the Properties for the COM port for Original Prusa i3 MK3, there was a message that the device wasn't completely set up. (That wasn't the exact message, just the jest of it,) So, on the drivers tab, I clicked Update Drivers. It wanted a reboot of my machine. After it booted up, I was able to flash with the latest firmware.
So, try updating the drivers by clicking that button and letting it search the internet for a newer driver.
