If you have IKEA where you live, IKEA Skarsta tables get stacked with acrylic sheets on sides and some "fancy" IKEA doors and hinges to make the state-of-the-art enclosures.

X = 43 cm
Y=35cm.
z=37.5cm

This is without the screen attached. The screen would add 7.5cm .
I will be using two Lack tables so I will have to put the spool of filament on the top of the table.

Yes, that looks like a good setup and I like the idea of making it a little taller. I did add some LED lights and a Raspberry Pi with Octoprint which so far has worked well for me. Overall this seems to be a low cost way to enclose the printer.

I am making an enclosure based on the 'HotBox' design.

That design was really for the Taz 3D printers and includes a vent for the 'eBox' on the TAZ.

Does anyone know if the power supply on the Mk2S needs venting or if it needs a fan for cooling when it is in an enclosure.

I am installing two 140mm variable speed Noctua fans (one feeding in cool air and the other extracting hot air) for general ventilation but not sure if I need something specifically for cooling the Mk2S power supply box.

I mean, I think you answered your own question. With your fan setup you proposed, that is going to move WAY more air than would ever move through a normal room inside there.

Kevin failed to consider the print bed movement. I‘d add another 10cm or even a bit more to allow the print bed to fully move fore and aft.

Standard Lack table is 550mm x 550mm outside edges, this allows MK2S to fit inside and lets bed move fully forward and bed cables move fully backward without restriction - IF - the LCD is mounted outside the enclosure. There are many mounts for external LCD and there are also a couple of options for LCD mounts to fit the side rods so you can install the printer across the enclosure.

You also need to give thought to how and where you will mount your spool holder(s) and add enough height for that. My enclosures are 600mm high.
