I don't think this is presently a "thing". If it is please enlighten me. If it isn't, I guess I'm off to Github.

I frequently print small items that I would like to make into negative molds which then can be filled with various pourable resins. I would love a technique/feature in PS whereby the item is rendered as a negative volume within an encompassing positive shape. I recognize I would probably have to manually add "spru channels" for pouring and some geometries would not be possible. For instance, a "thru" hole would require a floating positive cylinder within the negative volume. Fortunately,  my items do not have such negative spaces to begin with, or at least with round holes, I could possibly add them by drilling, post removal.

Is this worth pursuing as a slicer feature? Other suggestions to achieve this? I'm ignoring shrinkage or expansion of various materials at this stage of discussion.

You can already use any model as a negative volume.  So what's stopping you already doing this ?

First attempt: Created a "new part" (block) of sufficient size to enclose the item. When added as a negative volume, the item was roughly 200% in size. As a check, I added as a positive volume. It was normal (100%) sized. Stumped.

cant help you there without a saved prusa slicer project file to see the issue you are having.
