Layer thickness correction in one layer

If I have an Stl file, made in Fusion360 from an object that is exactly 40 mm high, sliced ​​in Prusaslicer with a layer thickness of 0.3 mm, Prusaslicer calculates the height to 40.1 mm. (1x0.2mm + 133x 0.3mm).
That is not what I want, I do want to choose the 0.3 mm layer thickness for speed gain.

Is there a method to accomplish this in Prusaslicer.

What’s your first layer configured as ?

also you can use variable layer height near the top to allow finer layers If needed too.

I mentioned this in the post,  (1x0.2mm + 133x 0.3mm). First layer = 0.2, adviced by Prusa, (maby it is possible to change that a little).

I can only use the variable heights method after slicing the object, and that gives the total height of 40.1 mm.

Within this height I can use variable heights, but that doesn't help me.

Change your first layer height to 0.3 and see. You will need to post your project .3mf file for anything further as I can’t see why you are having this issue. Just save it, zip it up and attach it then we can actually slice it and preview it with the same settings you are using. 

The variable layer height allows finer graduation if an exact number of layers can’t produce a specific height so after you slice you can set it and then reslice to get a new possible height.

@neophyl has you on the right track. Variable Layer Heights (VLH) can get closer to your desired level of precision. What's not immediately obvious is what limits the range of heights VLH can use. VLH is constrained by the minimum and maximum layer heights specified in your printer settings:

If you adjust that minimum, you can get closer, although an exact match may still be tricky.

Not sure what you mean by "I can only use the variable heights method after slicing the object, and that gives the total height of 40.1 mm." Select the part in 3D editor view and you can work with VLH.

For an example of my "problem" see the attached zip file from my 40mm cube.

I have further explored the use of the variable height adjustment through a Youtube video. I was under the impression that these layer thicknesses could only be adjusted after the slicing process. I now know how to do this and I manage to set the height of my object to be sliced ​​exactly in Slicer.
It would be even easier to be able to adjust, for example, only the top layer in height in Prusaslicer.

You can adjust the top layer height. Just add a layer height modifier, make sure it covers only the top and then select layer height as a modifier option.  Once I’m back at my desktop I’ll download your file and adjust and repost to give an example but it’s not hard to do.

OK, attached is a project file that slices at 40mm high.  No variable layer height used.  I just added a normal Layer Height range modifier between 39.8 and 40mm and made the layer height 0.1 in that range. 
The only other change I made was to your Printer>Extruder > Max Layer Height settings.  You were slicing at 0.3mm layer height in Print settings but your extruder max height was set to 0.25 so that was limiting it.
