Do I need to enter Z offset into Printer Settings?
After using the Configuration in the MK3S+ I find my Z offset to be -1.320. Do I also need to enter this into the Printer Settings, General of Prusa Slicer (it is currently zero). I'm thinking not. If not, and it is stored in firmware of actual printer, why is this setting here?

No you dont. This value is stored in the printers firmware and assuming you are using a current firmware you can actually store and recall values for several different sheets.
The slicer is not just for Prusa Printers. Its used by many different pieces of hardware. As such its there for those that need it.
