I've occasionally used 'Add Part' in Prusaslicer 2.3.0 and it's been okay, but now I'm having a challenge trying to make holes in a body by using parts with infill/ layers/ perimeters set to zero. On the attached part, I first added a box 'hole' that is at an angle and a smaller cylinder 'hole', also at an angle. The box does what it's supposed to, but I can't get the cylinder to provide the required hole. If I add perimeters it will add material but setting infill/ layers/ perimeters to zero on the cylinder seems to have no effect on the sliced part. If I add a third box hole, that seems to have no effect either... it's as if only one hollow part is allowed. In the part tree on the slicer, the first part has a solid orange circle in the corner, where everything else has a plus sign - what does that mean? Different slicing profiles have any affect. Is this me missing something or is there an issue here?

First of all thank you for attaching the project file.  Without it I we could have spent days suggesting all sorts of things when the problem is very simple.  

Your generic box is set to type:Modifier, with perimeters, infill and top/bottom set to zero so acts like a boolean cut (mostly).  
Your generic cylinder though it has the same settings, is of type:Part.  As its a part that you have defined as zero for all those things, in effect it doesn't do anything.  Its an invisible part as far as slicing is concerned.  If you had any of the infill or shell types at non zero you would have seen it sticking up out of your model. 
If you right click the cylinder and change type to Modifier then it negates the original object like I think you expect it to.  Just like the box does.

I'm afraid without the project file there's not much we can do but guess.

You need to save the project (File>save as) to save a .3mf file.  Then take that file, zip it into an archive and attach it to a post here.
With the project we can see exactly what settings you have, parts, modifiers, etc.  There are so many interacting things that without you are just shooting in the dark.  Which is ok on really common stuff but for anything else not so much.

That's great... so simple! I changed the part type to modifier and they now both have orange dots. Thanks for your help!
