Slic3r Removed Hole

The Prusa version of Slic3r had been working fine for me until I tried to slice this last design. The design calls for a "1" hole in the middle of the block being printed. The software displays the "1" just fine, but after slicing, the "1" is gone, both in the Preview tab and the Layers tab.
(See attached pictures.)

Anyone ever seen anything like it? Is there some setting that might correct it?

For now, trying to slice it with Cura, but would love to be able to stick to one platform moving forward.

Usually such STLs are not valid solids. You may run them through a STL fixing service as
https://service.netfabb.com/login.php

I had a similar issue with a DSLR camera lens hood I wanted to print. I projected geometry onto the face of my model, and extruded it inwards. The slic3r software did not recognize that it was in fact a hole. I think what it did, was model the hole as a transparent solid, and sliced it that way.

Try checking what the STL looks like in another previewer, or open the STL in your modelling software. If the hole does not show up in the preview in your modeller, try remodelling your '1' the exact same way that you modelled the 'st'.

Meshmixer and meshlab are two good programs for evaluating (and fixing) .stl files.
