I don't understand why my print is failing to print on one part of the first layer

I need some help identifying this problem.

I have had my MK3S+ for 2 weeks.

This is the third time that this print has messed up in the same way. At first I thought maybe the filament was moist, so I dried it at 50*C for 5 hours. It has improved the situation but the photo attached is the most recent one. I even turned down the speed from default for the perimeters.

I have removed the nozzle and cleaned it (0.40mm), and readjusted the Live Z adjust, which seems OK and not far at all from where it was pre-clean (-1.100).

I am using prusament silver PLA (came with the printer) 215/60*C 

What could be causing this problem? Could it be that the filament is still too wet? The printer is in a tiny room with a constantly running dehumidifier but it is an old house.

I have Prusament Silver PLA and it prints very nice (on Mini+). PLA isn't that sensitive to moisture -I had it opened for over three months and it was still printable. Btw. I wouldn't dry PLA above 45°C.
As it seems your first layer doesn't stick to bed. The reason for that is, your nozzle is too high (you need to decrease Z-offset in calibration). One can see on photo, that single line (on first layer) almost has a circle shape in cross section. It needs to be squeezed a bit, so it becomes flat on top. To be more precise: first layer should be about 0.2mm thick.
And needles to say: no fingerprints on print sheet!

The bed is PEI coated steel. I usually clean it pretty generously with isopropanol. 

I gave it a good scrub with a soft sponge and some soapy water and that seems to have made a big difference. I just did a first layer test and it came out muuch better than before, so I think the problem must have been a bit of residue of something on the sheet. 

 

I just realised that the problems started most strongly when I received new "blue roll" paper towels and started using isopropanol instead of methanol. I will experiment with different cleaning methods but it is only a short walk to the sink so perhaps I'll stick with that.

I had the same issue

I had the same thing happen. About two weeks of prints working beautifully every time and then suddenly adhesion problems. The IPA does not cut the grease from your fingerprints as well as detergent. Happy printing.
