Not sure exactly what you are asking; but a simple test for the extruder is to mark spots on the filament, 95 mm, 100 mm and 105 mm above the filament guide. Then manually extrude 100.0 mm of filament. The 100 mm mark should now be at the guide.

The point regarding the extruder step rate is vital. Slic3r expects that the machine will accurately produce a set amount of filament when told to do so. Too much will result in blobs and other imperfections in the print. Too little will result in gaps and poor inter-layer adhesion.

Please refer to the printer documentation and/or resources in the 3D printing community for details on how best to calibrate a particular machine.

Can someone please help me find the suggested thread on how to carry out this calibration?

Yes. That calibration is not recommended for the Mk3. There are no unknown variables to calibrate.
Also, slic3r is not the same as slic3r PE (prusa edition).

thank you for your reply, I have read about this process you have mentioned and I think this is exactly what I need to do, however, technically I am unable to work out how to do this ?
Could you please give me some ideas on how to carry out this task ?

Neither. The manual you are reading is for Slic3r. This is not the right manual for Prusa printers, and prusa’s version of Slic3r (Slic3r PE). You should be reading prusa’s manuals.

As stated above, those steps are not required for the Prusa Mk3/S printers.

If you are concerned or are experiencing seriously bad prints -- the endgame for the calibration is to ensure the extruder clicks so many times and producers so much filament. The best verification is to simply manually extrude 100 mm of filament via the settings manual axis move menu, and ensure by measurement 100 mm was actually extruded. If you find there is a significant difference you can change the related setting in Slic3r PE.
