Hello,
i wanted to slice a lithphan but this happened:
Prusa Slicer 2.6.1 (with default settings) made a infill for the empty middle section and "copied" the outer Perimeter for the actually smooth inner surface. With the 2.5.2 Slicer and previous ones this does not happen.
Can you help me which settings are wrong.

Possibly a broken model or it could be your settings. Load the model and make sure you have the issue, then Please save a project from Prusa slicer using file>save project as. Take the resulting 3mf file and ZIP it up first then attach it here. Must be zipped or the forum won’t attach the file. 
With a saved project we can see not only the model but also all the settings you are using. It’s a snapshot so allows us to slice just like you are doing.

Without it’s basically just guesswork.

David you attached the obj file.  We need your Prusa Slicer PROJECT file.  Make sure PS is giving you the issue then use File>Save Project As from the Prusa Slicer File menu to save the .3mf file.  The 3mf contains all the settings from the profiles you are using as well as the model.  Then zip and attach that.  

I can load your .obj in, I have to resize it to fit the bed of the printer as it exceeds my printer size but once I do it slices perfectly fine using a standard print profile.  

I can take a guess as to what it is but I cant be sure without your project.  My guess would be Print Settings>Advanced>Slicing>Slicing Mode.  I'd guess you have that set to Close Holes.  If you have then the slicer is doing exactly what its supposed to do and closing all the holes making it a solid disc.

i checked my profile and the Print Settings>Advanced>Slicing>Slicing Mode but it is already in normal mode.

Yes.  Your model was auto repaired because it had over 20000 errors on it.  Even after the repair there are still open edges.  

If you go into Preferences>Gui there's an option to show Non Manifold edges on models.  If you turn that on then any non manifold edge will show in purple.  Your model has those. I can clearly see a purple line going from top to bottom on the inside.   

Non Manifold issues with models are the ones that usually lead to messed up slicing as the model isnt 'watertight' so the slicer doesnt know what is inside or outside of the volume.  So you get filled in areas or missing areas etc.

Fix your model and it will slice correctly.
