Can you change colors by layer with the MMU2?
Is the layer color change available for the MMU2? If I switch back to the single filament configuration the plus symbol appears on the layer preview, but when I switch to the MMU2 configuration the plus symbol next to the layer slider goes away. Is there a trick to this that I am missing?

No, color change by layer is a single material system. You don't have a purge tower and you select the color by hand. MMU mode works with parts. Not by layer. You can split your object into parts and assign the material channels to those parts and use the purge tower. There is no combined mode.
