You probably should go back through the manual called "3D Printing Handbook" as others have said, if not to answer this problem, but to answer problems you'll likely encounter in the future without reading it.
As for why this particular issue is happening. The MMU2S is an add-on attachment for the MK3S and MK3S+, so it's not the same as the pure MK3S & MK3S+ profile. The MMU2S allows for multi-material printing and requires different instructions to tell the printer how to feed the filament.

That profile was probably selected because it shows many extruders on the slicer screen. I'd expect another question in a few days "Why can't the other colors print on my MK3S+ printer: they show up in the slicer profile."

Why am I so sarcastic? I've been on this forum long enough to know the pattern.

Everyone has to start somewhere, there will always be people asking the same questions. It's almost a fact of life, not just 3D printing or electronics. I'd rather help build a welcoming space than one that scoffs at new users, even if they didn't know exactly what they were getting into.
 
What is somewhat unique to emerging technologies is the view of "if it's expensive it should just work". Sometimes people just don't know in what ways newer things have bugs and eccentricities / non-casual user friendly design interfaces. That will almost certainly lead to people asking "dumb questions" if only because they've been asked thousands of times and it seems so common to someone who's been here.
 
If you don't feel like answering you really don't have to.

Friends steered me to buying Prusa for a variety of reasons. They also steered me to using the defaults. To only branch out after I was comfortable using the defaults, or when the defaults failed to get the results I wanted.  After all these years, I still use the defaults: they really do work that well. Sure, a few items are changed, but the serious parts of the process, the defaults simply work as expected. 

Another thing I learned is that Prusa is a work in progress. The Mk3 is an iteration of an idea, not a fully developed product. It is a tinkerer's device: for the Early Adopter -- minus one.  Sure, it works out of the box - most of the time. But only if you use the defaults, and read the manual - however terse it is. That said, once past the novice stage, it can be more powerful because it is a tinkerer's device. Two sides of the same coin, I guess. 

Experience has proven my friends right. Prusa is a great value -- a lot of capability for small change -- but it doesn't really compare with the turn-key-ness of something that sells for $3,000 USD, or more.  Experience has also taught me that the defaults are there for a reason; that drifting from them is something that should not be done until you can predict the outcome (or are simply trying something new to see what happens). 

What I also found when first exploring and learning about 3D printing is that the subject is buried in lore and hidden pockets of knowledge. And for every question there are many answers: most missing critical information, or outright wrong. The blind leading the blind sort of thing. e.g., Why is 60c the common Prusa bed temp for PLA when other printers use a different bed temp? Why does PLA not stick to a print sheet with disperse finger oils when PETG sticks regardless of them? Why does some PETG stick like superglue yet other brands of PETG not stick as well? 

Though, I digress.

When a question comes along "I have a Mk3S but I chose to use the Mk3S+MMU profile - will it make a difference?" Well, you have to ask questions in return to explore the knowledge level of the person asking the question. e.g., Does this person understand that selecting a profile for an Epson ink jet printer might cause issues if really printing to an HP laser printer?  Then, and only then, can a proper response be formulated. So "RTFM" is a proper response when you understand the person has never opened that book nor has a clue about what those profiles actually are.

I have yet to find a true turnkey 3D printer.  I have many and none are turnkey.

Wow, everyone seems particularly cranky today.

That being said, the error message and symptom seem pretty straight forward.

When you installed PrusaSlicer did you run the setup wizard that offered different printer configurations to install?

You should click only the printers you have when running the wizard.

What you have missed Henry is a quite a number of fruitless threads from the same OP.  This group as always dove in with some great suggestions - but the OP doesn't seem to be able to read - and just keeps asking the same questions and complaining about why nobody is helping him solve his problem.  I think this thread was yet another one of those - but the well  has been somewhat spoiled as to this OP.  Some people cannot be helped.

1) You can lead a horse to water, and most will drink.
